Product Overview
The Human IgG ELISA Kit is a sensitive and precise assay designed for the quantitative determination of human IgG levels produced in vivo and for evaluating immunoglobulin secretion by hybridomas in vitro.
This assay uses the antibody sandwich principle. A microtiter plate, pre-coated with a capture antibody specific to the human IgG Fc region, has been blocked and stabilized to serve as the solid phase. Samples, standards, and controls are directly added to the wells. After unbound IgG is removed through washing, an HRP-conjugated Detection Antibody Solution is introduced, binding to the heavy and light chains of the captured human IgG. Following additional washes, a tetramethylbenzidine (TMB) Substrate Solution is added, producing a blue color proportional to the amount of bound IgG. The reaction is stopped using Stop Solution, turning the blue product to yellow, and the optical density (OD) is measured at 450 nm with a microtiter plate reader.
