Vascular endothelial growth factor (VEGF or VEGF-A), also known as vascular permeability factor (VPF), is a 45 kDa homodimeric, disulfide-linked glycoprotein produced by various cell types that plays a key role in angiogenesis and vasculogenesis.

The VEGF family consists of VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E (Orf-VEGF), and placental growth factor (PIGF). Among the multiple isoforms of VEGF-A produced via alternative mRNA splicing from the VEGFA gene, VEGF165 is the most abundant and essential for angiogenesis.

VEGF governs several biological processes, including the proliferation and survival of endothelial cells, vascular permeability, macrophage cell migration, wound healing, bone formation, and the development of embryonic vasculature. VEGF-A interacts with receptors VEGFR-1 and VEGFR-2 to regulate angiogenesis and vascular permeability, while VEGF-C and VEGF-D primarily bind VEGFR-3 to control lymphangiogenesis. VEGF expression is stimulated by hypoxia and cytokines such as IL-1, IL-6, IL-8, and TNF-alpha.
